pPICα A, B, and C vectors are 3.6 kb vectors for expression and secretion of recombinant proteins in Pichia pastoris. The recombinant protein was expressed as an N-peptide fusion encoding the Saccharomyces cerevisiae á-factor secretion signal. These vectors enable high-level, methanol-inducible expression of relevant genes in P. pastoris and can be used with any P. pastoris strain, including X-33, SMD1168H, and KM71H. The pPICα vector contains the following elements:
• Contains the AOX1 promoter for tightly regulated methanol-induced expression of the relevant genes
• Provides all three reading frames (A, B, C version) to facilitate in-frame cloning using C-terminal peptides
• α-Factor secretion signal for directed expression to direct recombinant protein secretion
• Zeocin resistance genes for use in E. coli and Pichia pastoris Selection
• C-terminal peptide containing c-myc epitope and polyhistidine (6xHis) tag for detection and purification of recombinant fusion proteins
